Title: Paracrine regulation of endothelial cell motility and angiogenesis by deoxyribose-1-phosphate.

The regulation of angiogenesis is complex and involves the interplay of different molecular pathways. Its importance spans from cancer biology to regenerative medicine. Previous studies showed that thymidine phosphorylase (TP) and its enzymatic product deoxyribose-1-phosphate (dRP) stimulate angiogenesis. Although the biochemical and cellular mechanisms underlying the pro-angiogenic activity of TP and dRP are largely unknown, we proved that platelet-derived dRP promotes endothelial cell motility by modulating integrin expression in a redox-dependent manner and stimulates angiogenesis in vivo (Arterioscler Thromb Vasc Biol., 2010; 30(12):2631-8.). This project will elucidate the molecular mechanisms underlying this novel and important pro-angiogenic pathway, will advance our understanding of redox-dependent intercellular communication, and might represent the basis for developing regenerative medicine treatments based on dRP or its derivatives.

The specific objectives of this project will be: 1)to determine which endothelial cell structures and proteins interact with dRP using a proteomics approach; 2)to identify the molecular mechanism of dRP-dependent increase in endothelial reactive oxygen species (ROS) generation; 3)to define the intracellular events mediating the dRP-induced stimulation of endothelial cell motility downstream ofROSgeneration; 4)to assess whether the release of dRP by platelets stimulates angiogenesis in the mammalian cardiovascular system in aROS-, integrin β3-, and HO-1-dependent manner.

This project will be highly interdisciplinary and will suit a candidate with a background in Biochemistry and Cell Biology, Biomedical Sciences, Physiology or Pharmacology. Several state-of-the-art biochemistry, cell biology, proteomics, microscopy, and in vivo techniques will be used throughout the project. Strong interactions with several members of the Centre for Regenerative Medicine (CRM) of the University of Bath and external collaborators will guarantee invaluable learning and career-advancing opportunities. Because of the solid academic reputation of the Pharmacy and Pharmacology Department and its tight links with pharmaceutical industries, this position will represent an important step towards a successful career in cardiovascular research.
